Molecular cloning (basic course)
Who is invited?
The course is dedicated to employees of universities, research institutes, inspections, companies from the R&D sector, as well as students of medical or natural sciences who want to gain knowledge and expand their research workshop with practical tools from the field of molecular biology.
Programme
The course will familiarize participants with the methods of DNA cloning. Both theoretical and practical knowledge of DNA isolation techniques, polymerase chain reaction (PCR), methods of cloning genes into plasmid vectors will be gained during this course. The detailed programme:
Day I
- Introduction to molecular cloning (basic concepts: vectors, restriction enzymes, ligation, transformation. Discussion of various cloning techniques, e.g. restriction cloning, cloning due to homologous recombination, ligation cloning).
- Preparation of the insert for cloning (PCR reaction, purification of the amplicon).
- Digestion of plasmid DNA with restriction endonucleases, agarose gel electrophoresis, isolation and purification of DNA from the agarose gel.
- Obtaining a recombinant DNA molecule (ligation of the insert into the vector).
- Introduction of the recombinant DNA molecule into a bacterial cell: transformation of competent cells. Plating of bacteria on selective media, incubation of the cultures.
Day II
- Selection of the transformants.
- Colony PCR, agarose electrophoresis to confirm the presence of the insert.
- Isolation and purification of plasmid DNA. Assessment of the quality of the isolated genetic material.
- Preparation of plasmid for sequencing.
- Analysis of the results of plasmid sequencing (obtaining a consensus sequence, comparing results with sequences available in the NCBI database).
- Summary and discussion (discussion of the results, tips for troubleshooting and further experiments of the participants, most common errors, question and answer session).
General Information
The training will be conducted in small groups (up to 8 people) in the Research & Development Laboratory of IFB UG & MUG (20 Podwale Przedmiejskie street, 80-824 Gdańsk, Poland). Each participant will conduct all the experiments on his/her own in order to obtain the necessary laboratory practice. The courses will be conducted by experienced academia teachers, specialists and practitions in the field. Each day two coffee breaks and a lunch shall be provided. The participants will receive certificates confirming acquisition of the practical skills.
